2. Mirsky's fixative (National Diagnostics, Hull, Humberside, UK): Make from 10X concentrate and 10X buffer in distilled water. Store at 4°C for up to 1 mo.

2.3.1. p-Galactosidase Staining

1. X-gal stain solution (Subheading 2.1., item 8).

2. Bluo-gal stain solution. PBS (pH 7.3) containing 5 mM K3Fe(CN)6, 5 mM K4Fe(CN)63H2O, 1 mM MgCl2, 150 mM NaCl and 0.4 mg/mL Bluo-gal (5-bromo-3-indolyl-P-d-galactoside, Sigma). Store at 4°C in the dark. Bluo-gal can be made up as a 100X stock solution (40 mg/mL in N,N-dimethylformamide— store in dark at -20°C).
