1. Prepare the PCR mix (see Subheading 2.1., item 1) and incubate at 95°C for 5 min.
2. Incubate the PCR mix at 70°C while adding 2 pL (1.25 U) of Taq DNA polymerase.
3. Incubate the sample at 55°C for 1 min, then 72°C for 2 min, followed by 94°C for 1 min, and repeat for 35 cycles on a Hybaid PCR unit.
4. Incubate the sample for the final cycle at 72°C for 10 min and then at 20°C for 10 min before storing at -20°C. The labeled probe is stable for up to 12 mo at -20°C.
