Histology

Sections can be made of stained embryos by embedding in Durcupan (Sigma).

1. Fish embryos are incubated in a 1:1 mixture of xylene and Durcupan (Sigma) overnight at 4°C.

2. After evaporation of the xylene at room temperature in a fume cupboard, the embryos are embedded in fresh Durcupan.

5. Cut 2-3 pm sections with a microtome equipped with a glass knife (see Note 28).
