1. Embryos are dechorionated and stained in a solution containing 2 mM FDG and 20% DMSO in ddH2O for exactly 2 min and 30 s at 33°C. Note that older embryos that have muscular movements may be anesthetized in tricaine to aid viewing and photography.
2. Wash extensively with fish water.
3. Examine with a low-power objective (5X) on a fluorescence microscope with an FITC filter set.
4. False positives can develop after 30-40 min. Some tacZ-expressing embryos may be overlooked if expression is not very strong.
