Reagents

• Positive control: SAF-fixed faeces containing trophozoites of any species.

• Wheatley's (1951) trichrome stain: chromotrope 2R 0.6g, light green SF 0.3g, phospho-tungstic acid 0.75g, glacial acetic acid 1ml, distilled water 100ml. Add acetic acid to dry components. Allow to stand 15-30min at RT. Add the distilled water. Store at RT. The stain has a shelf life of 24 months.

• Acid-alcohol: 0.45% glacial acetic acid in 90% ethanol.

• Xylene or substitute, for example, Histoclear (National Diagnostics).
