1 Place a drop of 0.9% saline on one end of a microscope slide, on the other a drop of d'Antoni's iodine solution.

2 Using an applicator stick, pick up a small amount of specimen and mix thoroughly with the saline. Repeat the process this time mixing with the iodine.

3 Place a coverglass on each suspension.

4 Scan both preparations with the 10* objective, the saline suspension first. The whole coverslip area should be examined. When necessary, change to the 40* objective for a more detailed study. Some of the coverslip area always should be examined with this objective.
