The Cost Benefit Analysis of Polymorphisms within Lipoprotein Metabolism

Mthfr C677t Gene Polymorphism

APO-E4 is not the only thrifty allele to influence cholesterol levels. Ancestral diets rich in fiber, vegetable protein, plant sterols, etc., were clearly low in trans-fatty acids and the other substrates of cholesterol biosynthesis (Figure 2.10). It has been suggested that, as a result, a range of cholesterol raising polymorphisms exist that have been conserved by evolution (61). These include the B2B2 genotype of the cholesteryl ester transfer protein-TaqlB, which favors cholesterol transfer to the liver for bile acid synthesis (62), and the T54 GA AA genotypes of the intestinal fatty acid binding protein gene which favors fatty acid absorption for cholesterol synthesis (63). Clearly, a partial return to our ancestral diet of vegetables, fruit, and nuts would be an effective measure to reduce aberrant blood lipid profiles. Figure 2.10 shows how plant sterols like -sitosterol are structurally similar to cholesterol and hence may competitively reduce cholesterol absorption.

Thirsty Genes Putative Thrifty Genes that May Have Been Selected to Counter Water Stress but Now Lead to Hypertension

Intestine Cholesterol Absorption Gene

Humans have a physiological requirement for salt and mineral sodium. Indeed, one of our most basic senses of taste is for saltiness. This taste is not unpleasant, and it is perhaps unsurprising that in the West, our salt intake far exceeds our physiologic needs. Even the U.S. Food and Drug Administration recommendation of 6 g day for a low-salt diet is estimated to be 3.5x that of the typical Paleolithic diet. Some people are sensitive to salt and possess a genetic predisposition toward developing life-threatening hypertension (high blood pressure). Ten percent of people are affected by salt sensitivity in this way. Figure 2.10. The structural similarity between plant sterols like p -sitosterol and cholesterol results in molecular competition for intestinal absorption. Ancestral diets were rich in plant sterols and, hence, lower in cholesterol and its precursors. Figure 2.10. The structural similarity between plant sterols like p -sitosterol and cholesterol results in molecular...

Advantage of the Alanine Glyoxylate Aminotransferase AGT Pro11Leu Polymorphism in Meat Eaters

Activity along with a small amount of protein mistargeted between organelles. The effect of this on the synthesis and excretion of oxalate and deposition of calcium oxalate crystals in the kidney and urinary tract is thought to be significant. It has been postulated that there is a distinct advantage to possession of this SNP for populations that are predominantly meat eaters. It is believed that redirection of a small proportion (ca. 5 ) of AGT from peroxisomes to mitochondria in humans might lead to a subcellular partitioning of AGT that was better suited to an omnivorous as opposed to a herbivorous lifestyle (52) and, hence, offer meat eaters a distinct survival advantage. Caldwell et al. (53) recently showed that the frequency of Pro11Leu allelic frequency varied widely among 11 extant human populations with differing ancestral diets. The frequency varied from 27.9 in the Saami with a meat-rich ancestral diet to 2.3 in Chinese who have a more mixed ancestral diet. Using the...

Brief History Of The Human Diet

Early man met his needs for macro- and micronutrients via a largely herbivorous diet (12). It has been postulated that as man moved toward a more nutrient-dense diet, this facilitated an increase in brain dimension with a concomitant decrease in gut size (13). Such a diet was likely to contain not just animal fat and protein but plants (leaves shoots roots berries). Clearly, a significant mismatch exists between our ancestral diet and our contemporary one, which is energy rich-nutrient poor. This book examines not just some of the more interesting nutritional selection pressures that forged our species, and that are dealt with in this chapter, but it will also attempt to examine how incompatibility between programming in our ancestral genes and the dietary consequences of an agricultural revolution has led to the evolution of chronic debilitating human diseases within modern society (Figure 2.1).
