Abstract

Natural killer (NK) cells are a major component of the immune system, which can play an important role in host defences against cancer and microbial infections. Their identification, characterization, elucidation of how they function and are regulated, exploration of their biological roles have all occurred during the past 30 years. NK cells have been found to be distinct from T or B lymphocytes, have a characteristic morphology of large granular lymphocytes, and can be readily identified by characteristic cell surface molecules. The hallmark of their function is the spontaneous ability to reorganize and rapidly kill a wide array of tumour cells and also virus-infected normal cells. In addition, their cytotoxic activity can be strongly augmented by interferon, interleukin-2, and other cytotkines. NK cells not only respond to various cytokines, but are also major producers of some cytokines, especially interferon gamma. NK cells have been found to play a major role in host resistance against tumour growth and to be particularly effective in resistance against metastatic spread of tumours. Their potential to inhibit tumour growth has led to an increasing interest in exploring the therapeutic effects of activated NK cells against various types of cancer.
