Segregation analysis is used to determine the mode of genetic transmission of a disorder by describing the distribution of phenotypes in affected families. The two principal hypotheses which have emerged from these complex analyses in affective disorders are the single major locus model and the polygenic model involving possible interaction between two or more loci. The exact mode of inheritance is not yet clear. (20> The situation is further complicated by the possible presence of anticipation in families with mood disorder.(!.,2 and 3) This mode of transmission, which involves dynamic mutations, fits better with the twin and family epidemiological data available in affective disorders and may explain their non-Mendelian pattern of inheritance.
