The effects of NO on hearts are dose-dependent. Lower doses (nM) such as NO produced by eNOS and nNOS are beneficial to the heart (positive inotropic effects), but high doses (mM) such as NO produced by iNOS are harmful to the heart (negative inotropic effects).2734-37 Without the restriction of calcium calmodulin, iNOS produces larger amounts of NO and its activity continues.5 The molecular mechanisms of high levels of NO in the heart can be simply described as cGMP-dependent and cGMP-independent pathways.
