DNA and RNA am synthesized by adding each new nucleotide triphosphate to the 3' end of the growing polynucleotide chain (often referred to as synthesis in the 5' —» 3' direction).

What is the order of synthesis of a growing polypeptide chain? This was first determined in a classic experiment performed by Dintzis that was described in Chapter 2. This experiment found that each new amino acid must be added to the C-terminus of the growing polypeptide chain (often referred to as synthesis in the N- to C-terminal direction). As described in the next section, this directionality is a direct result of the chemistry of protein synthesis.
