Macrophages

When Cn spores are first inhaled, the host cells that initially phagocytose the yeast are presumably alveolar macrophages with neutrophil and dendritic cell involvement shortly thereafter. These important innate antigen-presenting cells are critical to initiating an effective immune response. Macrophages are thought to be one of the primary defenses against Cn infection. They help control the immunomodulatory effects of Cn by sequestering shed polysaccharide (GXM) in a receptor-dependent manner through phagocytosis or pinocytosis (He et al., 2003; Chang et al., 2006). However, the efficiency of phagocytosis is dependent on the cell cycle (Luo et al., 2006) and the activation state of the macrophage (He et al., 2003). In addition, a recent study found that macrophages derived from the B-1 subset of B lymphocytes show increased fungal killing via a NO-mediated mechanism (Ghosn et al., 2006).
